About the Job
Summary of Position
Perform and troubleshoot experiments and analyze data in a molecular biology and protein biochemistry / protein engineering laboratory. Maintain detailed scientific records. Collaborate with the Research Team Scientists to meet Laboratory Objectives and Key Results (OKRs).
Perform all activities with minimal supervision (weekly meetings with supervisor to review progress but day to day activities performed independently) in accordance with standard operating procedures. Maintain all associated laboratory equipment and facilities for the continued performance of these duties.
30% Molecular Biology and Cell Culture
Execute and troubleshoot complex molecular biology work flows including Gibson assembly-based sub-cloning and multi-fragment assembly, PCR, Sanger sequencing of plasmids, designing cloning strategies, phage-display and yeast-display-based molecular engineering.
Perform all activities with minimal supervision (weekly meetings with supervisor to review progress but day to day activities performed independently) in accordance with standard operating procedures. Maintain all associated laboratory equipment and facilities for the continued performance of these duties.
30% Protein biochemical and biophysical assays
Execute and troubleshoot complex protein purification work flows affinity purification of recombinant proteins, size-exclusion chromatography of recombinant proteins, and ion-exchange chromatography of recombinant proteins. Validate protein identity by mass-spectrometry and perform functional assays on purified proteins including (for example) enzyme-activity assays, protein-ligand binding assays, and stopped-flow-based measurements of biochemical reaction kinetics.
Perform all activities with minimal supervision (weekly meetings with supervisor to review progress but day to day activities performed independently) in accordance with standard operating procedures. Maintain all associated laboratory equipment and facilities for the continued performance of these duties.
30% Recombinant Protein Expression and Purification
Perform cell culture, developing, maintaining, and validating cell lines and strains including maintaining and culturing HEK cells in a dedicated BSL2 cell culture facility.
Perform all activities with minimal supervision (weekly meetings with supervisor to review progress but day to day activities performed independently) in accordance with standard operating procedures. Maintain all associated laboratory equipment and facilities for the continued performance of these duties.
